Description
The tCloneTet+PDB plasmid was obtained from part J100255 and was modified using Golden Gate Assembly to have a P14 promoter rather than the P5 promoter. The P14 promoter is known to have a lower efficiency than the P5 and the P2 promoter but has a higher efficiency than the P9 and P4 promoters.Notes
Used PCR to "cut out" the P5 promoter since there were no restriction enzyme sites surrounding it.Source
